Different effects of GsMTx4 on nocturia associated with the circadian clock and Piezo1 expression in mice

Nocturia is a major problem in geriatric patients. Clock genes regulate circadian bladder function and Piezo type mechanosensitive ion channel component 1 (Piezo1) that senses fullness. We utilized WT mutant (ClockΔ19/Δ19: nocturia phenotype) mice to determine if the effects of GsMTx4, Piezo1 inhibitor, dependent on expression bladder. compared voiding behavior after administration vehicle, low dose, or high dose GsMTx4. Intraperitoneal injections (IP) were performed at Zeitgeber time (ZT) 0, lower phase (ZT0-IP) ZT12, higher (ZT12-IP). Urine volume (Uvol), frequency (VF), urine per void (Uvol/v) measured using metabolic cages. VF decreased ZT12-IP only with GsMTx4 but showed no ClockΔ19/Δ19 mice. significantly ZT0-IP both doses, Uvol/v increased doses dose. did not affect Uvol ZT12-IP. A decrease was observed ZT0-IP; however, it unrelated GsMTx4-IP. The changed associated clock level. maximum effect occurred during sleep WT. These results may lead new therapeutic strategies against nocturia.
